DIOS Plates

AP/MALDI experiments can be performed on DIOS plates. Invented and patented by Dr. Gary Siuzdak, of the Scripps Center for Mass Spectrometry, DIOS targets are produced from crystalline silicon using specialized, proprietary etching procedures. DIOS plates are available from Waters (MassPrep DIOS plate part no. 186002322 for 1/pack and part no. 186002423 for 10/pack) and from Gary Siuzdak, The Scripps Research Institute Center for Mass Spectrometry (BCC-007), 10550 North Torrey Pines Road, La Jolla, CA 92037, USA (Part No. DS-100). A necessary DIOS plate holder for AP/MALDI ion source is accessible from MassTech, Inc (Part No. D96).

Typical Spectra:

MassPREP DIOS-target plate was washed with clean methanol and then allowed to air dry. The plate was loaded with 0.5 uL of Reserpine on to the spot, then the liquid spot was “dragged” to encompass the entire well. After drying, the plate was subjected to AP-MALDI PDF source interfaced with Thermo Finnigan LCQ Deca XP mass spectrometer. AP-MALDI works analogous to vacuum MALDI for the DIOS-target by Waters ™. Sample loading as per the instructions in the following spectra:

MassPREP ™ DIOS-target ™ Plate – Reserpine, FW (608.68), loaded 500 pg (800 fmol) Reserpine, FW (608.68), loaded 500 pg (0.8 pmol)

MassPREP™ DIOS-target™ Plate – fragments of m/z 609 (with 500 pg loaded) fragments of m/z 609 (with 500 pg loaded)

MassPREP™ DIOS-target™ Plate – loaded 100 pg (164 fmol) loaded 100 pg (164 fmol)
